Book Description: Tumor metastasis is usually associated with a poor prognosis for cancer patients. Notably, this pathological process is correlated with elevated expression and phosphorylation of f & barbelow;ocal a & barbelow;dhesion k & barbelow;inase (FAK), a non-receptor tyrosine kinase whose activity is regulated by the adhesive state of a cell. In order to metastasize, neoplastic cells must invade into the surrounding tissues and into the vascular systems. Tumor cell invasion is a complex process that generally involves the degradation of the surrounding e & barbelow;xtrac & barbelow;ellular m & barbelow;atrices (ECMs) followed by directional cell movement. To explore the molecular mechanism underlying the potential function of FAK in cell invasion, we generated transformed cell lines by stable expression of v-Src in FAK deficient (FAK-/- ) and wild type (FAK+/+) cells. Interestingly, we found that FAK played a central role in invasion through inhibiting endocytosis of a key cell-surface proteinase, MT1-MMP. We identified an interaction between FAK and endophilin A2 and showed that this interaction allowed phosphorylation of endophilin A2 at Tyr315 by a FAK/Src complex. This phosphorylation event disrupts the endophilin/dynamin complex that plays an essential role in endocytosis, thus attenuating endocytosis of MT1-MMP. In this way, FAK upregulates surface expression of MTI-MMP and consequent ability of cells to degrade the ECMs. In addition to regulation of ECM dissolution, FAK has also been implicated in controlling cell migration. We found that FAK associated and phosphorylated N-WASP at Tyr256. Our results also indicated that N-WASP localized in both the cytoplasm and the nucleus. Nuclear translocation of N-WASP is dependent on its being in the open conformation and could serve as a negative feedback loop to tune down its cytoplasmic function. Phosphorylation of Tyr256 of N-WASP could sustain N-WASP activation in the cytoplasm by inhibiting its nuclear translocation, thus facilitating cell migration. The nuclear localization of N-WASP inspired us to further analyze its potential functions in the nucleus. We found that nuclear N-WASP is within a large protein complex containing PSF/NonO, nuclear actin, and RNA polymerase II. Moreover, we presented evidence to elucidate a fundamental role of N-WASP in RNA polymerase II-dependent transcription.
